Police say two people were seen pushing a car onto train tracks on Adelaide's Seaford rail line before a train crashed into the sedan at Lonsdale on Monday night.
Services on the line were suspended after the passenger train struck the empty vehicle.
Police arrested two men, aged 21 and 23, from Christies Downs.
They were both charged with endangering life, illegal interference and property damage.
The men have been refused police bail and are expected to appear in the Christies Beach Magistrates Court today.
The unusual incident happened just after 7:00pm, along the station platforms.
Transport Minister Stephan Knoll said a review of the design of the train station was now warranted.
Unusually, the station platform is at the same level at as the carpark it is immediately next to.
"We'll be having a look at what modifications need to happen to the station design to make sure that we've got it right and we do minimise these issues happening into the future," Mr Knoll said.
Police said there was no-one inside the car, and the train hit the vehicle at a low speed.
They said it took several hours for emergency services to clear the tracks.
"Police have reviewed CCTV and two suspects were seen pushing the car towards the tracks before running off," SA Police said in a statement.
